It's not the size that matters but what you do with it. DNA sequence principles in the era of genome-wide datasets

Genome-wide data is being generated at an alarming rate. But what does it all mean? In this talk I will ponder on how DNA sequence principles can make sense of this evergrowing mass. I will elaborate on a specific class of genomic cis-regulatory elements, the Polycomb/Trithorax Response Elements (PREs), which in flies are bound by Polycomb group (PcG) and Trithorax group (TrxG) proteins and maintain the transcription states of several hundred developmentally important genes. PREs are essential to the correct identities of both stem cells and differentiated cells. We have previously developed a computational method enabling the prediction of PREs in fly genomes with high accuracy. Our aims are to identify common sequence elements underlying PcG recruitment in the mammalian genome and to apply PRE prediction to mammalian genomes as was done in flies.
