KRAS in pancreatic ductal adenocarcinoma: New diagnostic tools and mechanistic insigh

Abstract

The predominant histologic form of pancreatic cancer, ductal adenocarcinoma (PDAC), is a dismal disease with a poor patient outcome. There is a substantial need for progress in early detection and mechanistic understanding of the disease which may lead to better therapeutic approaches to improve patient survival. Pancreatic cancer is characterized by a nearly universal somatic mutation in the KRAS gene. Somatic KRAS mutations, despite having a pivotal role in the development of the disease, act in concert with additional genetic alterations or other pathophysiological conditions in order to cause pancreatic tumorigenesis. The primary objective of my PhD project is to explore the diagnostic potential of liquid biopsy and deepen our mechanistic understanding of pancreatic cancer, especially in the context of somatic KRAS mutations and associated risk factors, particularly obesity.
