Hormonal regulation of nuclear receptor coactivator GRIP1

Members of the SRC family of coactivators interact with many nuclear receptors such as the glucocorticoid, estrogen, retinoic acid, thyroid hormone and peroxisome proliferator-activated receptors. Coactivators such as the SRCs exhibit intrinsic histone acetyltransferase activity that lead to remodelling of chromatin and recruitment of the basal transcription machinery. The SRCs have similar and quite ubiquitous expression patterns. Signalling pathways controlling nuclear receptor coregulators may indirectly affect the transcriptional activity of a number of nuclear receptors and subsequently a variety of cellular functions. We have demonstrated that activation of the cAMP/PKA pathway modulates the function and the subcellular localization of coactivator GRIP1. This is in contrast to two other members of the SRC family, SRC-1 and pCIP. Our results may represent a novel regulatory mechanism whereby peptide hormones regulate a nuclear receptor coactivator.
