Bakgrunn

Omtrent 1/3 av alle kreftpasienter får spredning av kreften til hjernen (hjernemetastaser), og metastaser til hjernen er 10 ganger mer vanlig enn kreft som oppstår i hjernen. De mest vanlige krefttypene som sprer seg til hjernen, er lungekreft (50-60%), brystkreft (15-20%), hudkreft (5-10%), og kreft i mage/tarm (4-6%).

Kreftpasienter med hjernemetastaser har en dårlig prognose, og dagens kreftbehandling øker ikke pasientoverlevelsen nevneverdig. Det er derfor nødvendig med forskning for å kunne finne nye behandlingsmetoder, og i denne sammenheng er det behov for gode modeller for å kunne forstå metastaseringsprosessen bedre. 

Utvikling av nye dyremodeller for å studere kreftspredning til hjernen

Vi injiserer kreftceller fra humane hjernemetastaser direkte inn i blodstrømmen på immundefekte mus. Vi ser da at cellene sprer seg til dyrehjernen, på samme måte som man observerer i pasienten. Disse modellene rekapitulerer de ulike stegene i kreftspredningsprosessen, og gjør oss i stand til å effektivt kunne teste ut nye behandlingsalternativer.

Pågående forskningsaktiviteter

Vår forskning fokuserer på 3 ulike angrepsmåter:

Vi har utført genetiske studier av kreftspredningen i våre dyremodeller, og bestemt en gensignatur for hjernemetastaser fra hudkreft. På bakgrunn av disse dataene har vi funnet flere ulike medikamenter, idag i bruk på andre sykdommer enn kreft, som viser seg å hemme veksten av hudkreft i hjernen.

Vi studerer blod-hjerne barrieren (BBB), som blokkerer medikamenter fra å trenge inn i hjernen, og dermed hindrer effektiv behandling. I et forskningsprosjekt har vi vært istand til å åpne BBB på forsøksdyr, og dermed oppnå god effekt av behandlingen.

For tiden studerer vi betydningen av såkalte exosomer i spredningen av hjernekreft. Vi har lovende data som viser at mikro-RNA levert til hjernen av exosomene, øker kreftspredningen. Vi har greidd å hemme denne spredningen ved å genetisk blokkere enkelte mikro-RNA til hjernen.  Dette er et pågående samarbeide med partnere ved Shandong University, Kina, og University of Luxembourg.
