The research group focuses on the structure, function and dynamics of myelin proteins. Important results have been produced on the structure and function of myelin proteins, as well as protein-membrane interactions and lipid membrane stacking. However, knowledge about molecular structure and interactions in a membrane with a natural myelin biochemical composition is limited.
This project will use structural biology and nanoscience to address the fine molecular details of how myelin proteins interact with each other and with model membranes. It will assess the mechanisms of lipid bilayer stacking by native-like combinations of CNS and PNS myelin proteins and will include both integral and peripheral membrane proteins that stack lipid bilayers spontaneously in vitro. This PhD project will answer questions regarding the effects of myelin proteins on stacking lipid membranes when they are combined, how they affect each other’s properties, how they reside within lipid assemblies, and whether they compete or function in synergy. Examination of structural differences will be conducted and compared to the structures of the individual proteins. Does the protein mixture reveal new properties relative to those already established? Mutated proteins will be used to see effects on stacking of bilayers in protein mixtures. This research will improve our understanding of myelin.
