Prosjekter

Gudbrandsen har ledet flere store prosjekter der formålet har vært å lære mer om hvorvidt inntak av fisk eller fiskeproteiner påvirker helsen til mennesker, og der overvektige rotter brukes som mekanistisk modell. De fleste prosjektene har vært gjennomført uten teknisk assistanse, men med utmerket bidrag fra masterstudenter og PhD-kandidater. Forskningsgruppen Proteingruppa, under ledelse av Gudbrandsen, ble etablert i januar 2015, for å bli mer synlig som forskningsgruppe og for å synliggjøre kompetansen vår. Nå er flere av prosjektene kommet så langt at vi kan starte publiseringsarbeidet, og vi forventer at 2016 og 2017 blir gode publiseringsår for Proteingruppa. Under er prosjekter som er under publisering, aktive eller under planlegging.

 

Fiskespisestudiene 2010-2014.  Prosjektleder: Gudbrandsen.

Finansiert av BMFS og støttet av industripartnerne Skretting ASA, Lerøy AS, BlueProtein, Nutrimar, Norsildmel, Bergen Teknologioverføring (BTO) og Halstensen Granit AS, i tillegg til støtte fra Regionalt Forskningsfond Vest til to av delstudiene (del 3 og 4; fiskeproteiner). Helseeffekter av inntak av fet og mager fisk, og proteiner isolert fra fet og mager fisk, ble undersøkt i normalvektige og overvektige. Fire delstudier: 1. Helseeffekter av høyt fiskeinntak i normalvektige voksne, 2: Helseeffekter av høyt fiskeinntak i overvektige voksne, 3: Helseeffekter av lavt inntak av proteiner fra sild, laks og torsk som supplement i overvektige voksne, 4. Helseeffekter av inntak av vannløselig og vannuløselig torskeproteiner i overvektige voksne. I tillegg ble tre rotteforsøk utført med fiskeproteiner; en artikkel er publisert (Drotningsvik 2014), og de neste artiklene sendes til tidsskrifter for vurdering i 2016. Syv manuskripter fra de kliniske studiene og et manuskript fra rottestudiene er under arbeid, av disse er et nå under revisjon i British Journal of Nutrition.

 

Korttidsstudien. Prosjektleder Gunnar Mellgren

Gudbrandsen har vært ansvarlig for koordinering og gjennomføring av studien. Samarbeid med Voss Sjukehus og Haugesund Sjukehus. Delvis finansiert av BMFS for analyser m.m. Studien sammenligner endringer etter to typer fedmeoperasjer; gastric sleeve (Voss) og gastriv bypass (Haugesund) en uke etter operasjon, med fokus på regulering av blodsukkeret. Avslutning av prosjektet har vært svært forsinket pga dødsfall (kirurgen i Haugesund) og to graviditeter, og fordi pasientene hadde problemer med å fullføre testene i studien pga oppkast, men nå er alle prøver tatt og prosjektene ved sykehusene er avsluttet. Analyser av prøver fra prosjektet er i gang, og første manuskript forventes ferdigstilt i løpet av våren 2016.

 

Fiskeproteinstudiene 2015-2019. Prosjektleder Gudbrandsen

Helseeffekter av fiskeproteiner i fysisk aktive og inaktive voksne, godkjent av REK (2015/75). Vi vil sammenligne effekter av fiskeproteininntak i eldre med sarkopeni, voksne med overvekt og voksne som trener styrke. Fokus vil være på inflammasjon, glukoseregulering, tarmbakterier, næringsopptak. Samarbeid med Vedde/TripleNine, K. Halstensen, Haukeland Universitetssykehus, Norsk Sjømatsenter, Kjemisk institutt UiB og NOFIMA. Forskningsrådet har støttet prosjektet med to nærings-PhDer, Aslaug Drotningsvik (01.01.15-31.12.18) og Iselin Vildmyren (01.10.15-30.09.18) med Gudbrandsen som hovedveileder og henholdsvis Ola Flesland (Vedde) og Christian Halstensen (K. Halstensen) som biveiledere. Samarbeid med Halstensen Granit As, Sjømatsenteret og NOFIMA. Pilotstudie inngikk under Fiskespisestudiene og ble delvis støttet av Regionalt Forskningsfond Vest (200 000 NOK). Hovedprosjektet har fått 3 mill NOK i støtte fra Regionalt Forskningsfond Vest og er tilknyttet en bevilgning fra Fiskeridirektoratet (880 000 NOK) for produktutvikling og utprøving av fiskeproteiner.

 

Fish Intervention Studies (FINS). Arbeidspakke 2 (Helseeffekter av fiskeinntak i overvektige voksne og barn) ledes av Gudbrandsen.

Samarbeid med bl.a. Haukeland Universitetssykehus, NIFES, Kjemisk institutt UiB, Norsk sjømatsenter, Lerøy AS og Skretting AGA, finansiert av Fiskeri- og Havbruksnæringens Forskningsfond (FHF) og Forskningsrådet. 300 voksne og 155 barn med overvekt skal inkluderes i denne diettintervensjonsstudien, der deltakerne skal endre spisemønsteret sitt mot sunnere middager med fisk eller andre proteinkilder i 12 uker. Studien er nå halvveis i voksenstudien, med 150 inkluderte. Første artikkel vil omhandle effekter av fiskeinntak på blodsukkerregulering i et utvalg av de overvektige voksne som har fullført intervensjonen, og er planlagt innsendt desember 2015. PhD-kandidat Linn Anja Vikøren er ansatt i dette prosjektet (01.09.13-31.08.16), med Gudbrandsen som hovedveileder. Totalt 70 mill NOK inngår i FINS-prosjektet.

 

Menn med mage. Prosjektleder Gunnar Mellgren

PhD-kandidat Vivian Veum, i samarbeid med Simon Dankel og Jørn Sagen. Gudbrandsen er biveileder for Veum og har deltatt i planlegging, analyser og tolkning av resultater. Middelaldrende menn med fedme har fulgt dietter med mye mettet fett eller lavfettdiett for å sammenligne effektene av disse diettene på risikofaktorer for hjertekarsykdom. Første artikkel fra prosjektet er innsendt til American Journal of Nutrition for vurdering.

 

Direkte pankreasfunksjonstesting for diagnostikk av pankreassykdomer. Prosjektleder Georg Dimcevski (Gastro)

Samarbeid med bl.a. PhD-kandidat Friedemann Erchinger, Dimcevski, Erling Tjora, Odd Helge Gilja, Trygve Hausken, Trond Engjom og Helge Ræder. Prosjektet søker å utvikle nye og raskere analysemetoder for diagnostisering av pankreassykdommer (kronisk pankreatitt, pankreasfunksjon ved diabetes, cystisk fibrose, pankreascancer) og malabsorpsjon ved måling av bikarbonat, amylase og lipase i duodenalsaft etter stimulering med sekretin. Gudbrandsen har nylig utviklet en automatisert bikarbonatmåling som nå er godkjent for bruk i klinikken, og det arbeides for godkjenning for automatiserte metoder for amylase og lipase for bruk i klinisk rutine i løpet av 2015. To postere har blitt presentert fra dette arbeidet (Erchinger 2014, Aasmul 2014). Første artikkel (Automated spectrophotometric bicarbonate analysis in duodenal juice compared to the back titration method ) er publisert i Pancreatology (2016). Ytterligere to artikler er under arbeid og er planlagt ferdig i løpet av våren 2016, og disse tre artiklene vil inngå i Erchingers PhD-avhandling.

 

Fluid overload and motility disturbances in the small intestine - characterization of the effects of an activating GUCY2C mutation. Prosjektleder Torunn Fiskerstrand

Samarbeid med bl.a. PhD-kandidat Hilde von Volkman og Fiskerstrand. Prosjektet vil karakterisere fenotypen til pasienter med mutasjon i GUCY2C, som kjennetegnes ved diare/løs avføring og liten tarmmotilitet. Gudbrandsen deltar i analysearbeidet av blodprøver for hormoner og lipopolysakkarider, og optimatiserer metode for analyse av serotonin i blodprøver fra disse pasientene.
