Abstract
Parkinson’s disease (PD) therapies that halt or slow disease progression are urgently needed. This project targets mitochondrial complex I deficiency, a key feature in a subgroup of idiopathic PD, using an innovative cell-based drug-repurposing strategy. Screening of CNS-penetrant compounds identified a small-molecule compound (Hit1) as a promising candidate that increases mitochondrial respiratory chain protein levels, improves oxygen consumption, rescues rotenone-induced mitochondrial fragmentation, and stimulates mitochondrial biogenesis. These findings support further validation of Hit1 and related analogues as potential disease-modifying therapies for PD, with broader relevance for mitochondrial disorders in both the brain and peripheral tissues.
