Not alone: protein-protein interactions between ion channels and endogenous or de novo designed protein binders

Membrane proteins such as ion channels play crucial roles in human physiology and their dysfunction is associated with a variety of diseases, such as neurological disorders and cancer. Despite recent advances in our understanding of their structure and function, many aspects of their biology remain elusive. In particular, the consequences of protein-protein interactions on their function are poorly understood. Here, I will present work in which we have investigated the regulation of acid-sensing ion channels and the NALCN sodium leak channelosome by protein-protein interactions, mediated by either endogenously expressed proteins or de novo designed miniprotein binders.
